External NADH oxidase type 2 (Nox-2 or NDH-2) is an enzyme that catalyzes the oxidation of NADH by transferring electrons to molecular oxygen or quinones. This process regenerates NAD(+) from NADH, which is essential for cellular redox balance and energy metabolism. It is located on the external face of the mitochondrial inner membrane in eukaryotes and is membrane-associated in prokaryotes. Unlike Nox-1, Nox-2 primarily forms water as a product and is resistant to complex I inhibitors like rotenone, capsaicin and piericidin A. Nox-2 plays a critical role in maintaining the cellular pool of oxidized nicotinamide adenine dinucleotide (NAD(+)).
