Overview

Extra domain A-containing fibronectin (EDA-FN) is a splice variant of fibronectin that is virtually absent from healthy adult tissues but highly expressed during periods of active tissue remodeling, such as embryogenesis, wound healing, fibrosis, and cancer (Source: UniProt P02751). It is a key component of the oncofetal extracellular matrix, where it promotes cell adhesion, migration, and the differentiation of fibroblasts into myofibroblasts through interactions with integrins (α4β1, α9β1) and Toll-like receptor 4 (TLR4) (Source: PubMed PMID: 24508223). Because of its high stability and restricted expression pattern, EDA-FN serves as an ideal target for the selective delivery of therapeutic agents. In oncology and chronic inflammation, EDA-FN is targeted by high-affinity antibodies like L19, which are used to deliver cytokines or radioactive isotopes specifically to the disease site, thereby enhancing therapeutic efficacy while minimizing systemic side effects (Source: PubMed PMID: 29153511).

Mechanism of action

Antibody-drug conjugates or immunocytokines (e.g., L19-based) selectively bind to the EDA domain of fibronectin in the neovasculature or stroma of tumors and inflammatory lesions, delivering therapeutic payloads like cytokines (TNF, IL-2, IL-12) or radionuclides directly to the disease site to induce local immune activation or cytotoxicity (Source: PubMed PMID: 29153511, 30243991).
