Overview

The Extra Domain A (EDA) of fibronectin, also known as EIIIA, is a protein domain inserted into the fibronectin molecule via alternative splicing of the primary RNA transcript (UniProt P02751). While virtually absent from normal adult tissues, EDA is highly expressed during periods of active tissue remodeling, such as embryonic development, wound healing, fibrosis, and tumor neo-angiogenesis (Schliemann et al., 2009, PMID: 19147758). It serves as a potent endogenous ligand for Toll-like receptor 4 (TLR4) and certain integrins, thereby promoting pro-inflammatory responses and cell proliferation (Gondokaryono et al., 2007, PMID: 17475861). In the context of drug development, EDA is utilized as a highly specific marker for the tumor microenvironment and inflammatory lesions. Therapeutic strategies often involve using high-affinity antibodies, such as the F8 clone, to deliver cytotoxic or immunomodulatory payloads directly to the site of disease, minimizing systemic side effects (Philogen Pipeline, 2024).
