Overview

Extracellular oncogenic receptors are a class of cell surface receptors whose aberrant (oncogenic) activity drives cancer progression. These receptors are typically located on the plasma membrane and can be activated by mutations, overexpression, or abnormal ligand binding. They play critical roles in transmitting signals from the extracellular environment into the cell, leading to altered cellular behaviors such as proliferation, survival, invasion, and metastasis. Common examples include receptor tyrosine kinases (RTKs) like EGFR and ERBB2/HER2, as well as G protein-coupled receptors (GPCRs), which can become oncogenic through mutation or dysregulation. These receptors regulate key processes such as cell growth, differentiation, migration, survival/apoptosis resistance. Oncogenic activation often results from mutations in extracellular domains leading to constitutive activation, overexpression/amplification, or abnormal ligand availability or autocrine/paracrine stimulation. They drive tumorigenesis by activating downstream pathways like MAPK/ERK and PI3K/AKT and contribute to hallmark cancer traits: sustained proliferative signaling, evasion of growth suppressors/apoptosis resistance, angiogenesis induction, invasion/metastasis. Tumor cells release EVs containing these oncogenic receptors or their fragments. EV-mediated transfer allows horizontal propagation of oncogenic signaling between cells—amplifying malignancy within tumors and potentially at metastatic sites.
