Extracellular signal-regulated kinase 8 (ERK8), also known as MAPK15, is a member of the mitogen-activated protein kinase (MAPK) family. It plays diverse roles in cellular processes, including cell adhesion, cell cycle control, and apoptosis. ERK8 uniquely interacts with nuclear receptors like ERRα, acting as a corepressor. It's implicated in protecting genomic integrity, regulating glycosylation, and influences organelle dynamics. Aberrant ERK8 function has been linked to cancers such as breast and colon cancer, suggesting a complex role as both an oncogene and tumor suppressor depending on the cellular context. Its unique structural features offer opportunities for selective small-molecule inhibitor development.
