The extracellular signal-regulated kinase (ERK) signaling pathway, also known as the MAPK/ERK pathway, is a conserved intracellular signaling cascade that transmits signals from extracellular stimuli to regulate diverse cellular processes. ERKs are serine/threonine protein kinases and are part of the mitogen-activated protein kinase (MAPK) family. The pathway is typically initiated by activation of receptor tyrosine kinases (RTKs) activating Ras which activates Raf kinases, which then phosphorylate and activate MEK1/2, which in turn phosphorylates ERK1/2.
