Overview

The ERK-SP1-MUC5AC signaling pathway is a critical regulatory axis involved in the production of mucus within the respiratory epithelium. In this pathway, extracellular signals such as growth factors or inflammatory cytokines activate the Extracellular Signal-Regulated Kinase (ERK) cascade, leading to the phosphorylation and activation of the transcription factor Specificity Protein 1 (SP1). Once activated, SP1 binds to the promoter region of the MUC5AC gene, driving the overexpression of Mucin 5AC, the primary gel-forming mucin in the airways. This pathway is frequently hyperactivated in chronic inflammatory lung diseases like asthma and COPD, where it contributes to mucus hypersecretion and airway obstruction. While not a single drug target itself, the individual components—particularly the ERK kinases—are major focal points for therapeutic intervention to alleviate respiratory distress and improve lung function.

Mechanism of action

Inhibition of ERK1/2 phosphorylation prevents the activation and nuclear translocation of the transcription factor SP1, which in turn reduces the transcriptional upregulation of the MUC5AC gene, thereby decreasing mucus hypersecretion.
