Overview

Ezrin (EZR) messenger RNA (mRNA) encodes a cytoplasmic peripheral membrane protein that functions as a critical linker between the plasma membrane and the actin cytoskeleton [UniProt: P15311]. As a member of the Ezrin-Radixin-Moesin (ERM) family, it is essential for maintaining cell shape, polarity, and the organization of cell surface structures such as microvilli [NCBI Gene: 7430]. In clinical oncology, EZR mRNA is recognized as a significant driver of metastasis; its overexpression is linked to the invasive phenotype of various malignancies, most notably osteosarcoma, breast, and lung cancers [PubMed: 15548694]. Therapeutic strategies targeting EZR mRNA, such as siRNA-mediated knockdown or antisense oligonucleotides, aim to reduce the synthesis of the ezrin protein to suppress tumor cell migration and invasion [PubMed: 22431513]. While small molecule inhibitors like NSC305787 target the protein directly, mRNA-based approaches offer a highly specific method for silencing the EZR gene's pro-metastatic activity. This target is particularly relevant in pediatric osteosarcoma, where high ezrin levels correlate with poor survival and early pulmonary metastasis. Downregulation of EZR mRNA has been shown to disrupt the interaction between the cytoskeleton and signaling molecules like RhoA, further inhibiting oncogenic signaling. Despite its potential, therapeutic development must account for the physiological role of ezrin in normal epithelial tissues to avoid systemic toxicity.
