Overview

F-actin-capping protein subunit alpha-1 (CAPZA1) is a cytoskeletal protein that regulates actin filament growth by binding to the fast-growing (barbed) ends of actin filaments in a calcium-independent manner, blocking subunit exchange and preventing further filament assembly. Unlike severing proteins such as gelsolin, CAPZA1 does not break filaments but acts to stabilize their length and organization. It is implicated in the formation of epithelial cell junctions and the regulation of molecular motor complexes like dynactin for intracellular transport. Mutations in CAPZA1 have been associated with certain muscular and kidney diseases, but it is not considered a current therapeutic target or biomarker.
