F-box and leucine-rich repeat protein 10 (FBXL10), also known as KDM2B or JHDM1B, is a substrate-recognition component of the SCF (SKP1-CUL1-F-box protein)-type E3 ubiquitin ligase complex with histone demethylase activity via its JmjC domain, catalyzing demethylation of H3K4 and H3K36. It contains an N-terminal JmjC domain, CXXC zinc finger, PHD finger, F-box domain, and leucine-rich repeats, enabling binding to CpG-rich promoters and interactions with Polycomb Repressive Complex 1 (PRC1) components like Ring1B for H2AK119 mono-ubiquitination. FBXL10 promotes cell proliferation, bypasses senescence by regulating EZH2 and H3K27me3, represses developmental genes, and is overexpressed in cancers such as acute myeloid leukemia, seminomas, and pancreatic ductal adenocarcinomas. Long isoform knockout leads to neural tube defects and perinatal mortality.
