Overview

F-box and leucine-rich repeat protein 15 (FBXL15) is a substrate recognition component of the SCF (SKP1-CUL1-F-box) E3 ubiquitin-protein ligase complex, responsible for targeting specific proteins for ubiquitination and proteasomal degradation[1][2][9]. FBXL15 specifically mediates the ubiquitination and subsequent degradation of target proteins such as Smurf1, Smurf2, and WWP2, thus acting as a positive regulator of the bone morphogenetic protein (BMP) signaling pathway[1][2][4]. Through this regulation, FBXL15 plays a critical role in embryonic dorsal/ventral pattern formation, maintenance of bone mass, and transitions in the cell cycle[2][1]. Disease associations include bone density defects when FBXL15 is dysregulated, and possible connections to inherited diseases such as Bardet-Biedl syndrome 10 and neuronal ceroid lipofuscinosis[1][9]. As of now, there are no drugs, clinical biomarkers, or established safety concerns specifically associated with FBXL15[1][2][9].
