Overview

F-box and WD repeat domain containing 12 (FBXW12) is a member of the F-box protein family, characterized by the presence of an approximately 40-amino acid F-box motif and WD repeat domains. It acts as a substrate-recognition component of the SCF (SKP1-CUL1-F-box protein)-type E3 ubiquitin ligase complex, facilitating the ubiquitin-mediated degradation of target proteins, which is crucial for the regulation of cellular processes such as cell cycle progression, growth, differentiation, and apoptosis[1][2][3]. The gene may play a role in cellular homeostasis, and its altered expression or loss, such as through genetic deletion or promoter methylation, has been associated with ovarian carcinogenesis and neuroblastoma[3]. Additionally, expression of a circular RNA from this locus (circ-FBXW12) promotes pathogenic cell behaviors in diabetic nephropathy, suggesting a function in both malignant and non-malignant disease contexts[3]. There are currently no known drugs that directly target FBXW12, and it is not established as an actionable therapeutic target.
