Overview

F-box only protein 10 (FBXO10) is a member of the F-box protein family, characterized by the presence of an approximately 40-amino acid F-box motif that enables interaction with the SCF (SKP1-CUL1-F-box protein) E3 ubiquitin ligase complex[1][5]. FBXO10 acts as a substrate-recognition component within SCF complexes, mediating the ubiquitination and degradation of specific protein targets. Notably, FBXO10 targets the antiapoptotic protein BCL2 for ubiquitination, thereby promoting its degradation and playing an important role in apoptosis regulation[1][5]. FBXO10 also participates in the ubiquitination and subsequent degradation of RAGE (receptor for advanced glycation end products) and the regulation of BCR signaling via degradation of HGAL/GCSAM[1]. Dysregulation of FBXO10 has been associated with lymphoid malignancies, particularly large B-cell lymphoma[1]. The broader FBX protein family, including FBXO10, is involved in critical cellular processes such as protein turnover, cell cycle regulation, and response to cellular stress[2]. While no drugs are currently known to specifically target FBXO10, its role in disease suggests potential future as a therapeutic target.
