F-box only protein 17 (FBXO17) is a member of the F-box protein family, functioning primarily as the substrate-recognition component of SCF (SKP1-CUL1-F-box protein)-type E3 ubiquitin ligase complexes. It recognizes denatured glycoproteins and sulfated glycans, directing them for ubiquitination and subsequent degradation. FBXO17 plays key roles in regulating type I interferon (IFN-I) signaling by binding IRF3 and facilitating its dephosphorylation via recruitment of protein phosphatase 2A (PP2A), acting as a negative regulator of the innate immune response. In oncology, FBXO17 is upregulated in several cancer types, including glioma and lung carcinoma, where it promotes cell proliferation and tumor progression by modulating Akt/GSK-3β/Snail and ERK/mTOR signaling pathways. FBXO17 is an emerging therapeutic target, serving as both a mechanistic node in cell signaling and a potential biomarker for cancer progression.
