Overview

F-box only protein 24 (FBXO24) is a member of the F-box protein family that serves as the substrate-recognition component of the SKP1–CUL1–F-box (SCF) E3 ubiquitin ligase complex, mediating K48-linked polyubiquitination and subsequent proteasomal degradation of specific target proteins, including nucleoside diphosphate kinase A (NDPK-A), PRMT6, and LSD1[1][2][3][5]. FBXO24 is prominently expressed in testicular cells, especially round and elongating spermatids, and plays a critical role in sperm development, particularly mitochondrial and flagellum structure, likely affecting male fertility[1][4]. In somatic cells, FBXO24 is implicated in chromatin regulation and suppression of tumorigenesis; its loss can promote cancer cell proliferation, EMT, and stemness via defective degradation of targets such as LSD1 and likely PRMT6[3]. Disease relevance includes its identification as a candidate tumor suppressor gene in diffuse gastric cancer, association with breast cancer, and links to chromosomal deletion syndromes; however, no therapeutic drugs are currently known to target FBXO24 directly[3][5].
