Overview

FBXO33 is an E3 ubiquitin ligase and a member of the F-box protein family that functions as the substrate recognition component of an SCF complex. It mediates the ubiquitination and proteasomal degradation of proteins, playing a role in post-translational regulation. In cancer, specifically gallbladder cancer, FBXO33 is upregulated and promotes metastasis and progression by driving EMT through the ubiquitination of p53, leading to its degradation. FBXO33 is regulated transcriptionally by YY1, which is activated via lactylation in the tumor microenvironment, reinforcing its expression and protumorigenic role. FBXO33 gene variations may additionally be linked with neurodevelopmental disorders and bone density.
