Overview

F-box only protein 43 (FBXO43), also known as EMI2, is a member of the F-box protein family, characterized by the presence of an F-box motif (~40 amino acids) that mediates protein-protein interactions within the SKP1-cullin-F-box (SCF) E3 ubiquitin ligase complex[3]. FBXO43 is involved in regulating the cell cycle, particularly through inhibition of the anaphase-promoting complex/cyclosome (APC/C), and plays a critical role in meiosis and early embryonic development. In somatic cells, FBXO43 is implicated in the regulation of cell proliferation, migration, and epithelial-mesenchymal transition. Its overexpression promotes tumor progression and is associated with poor prognosis in cancers such as hepatocellular carcinoma and breast cancer[1][2]. Mechanistically, FBXO43 interacts with and stabilizes CCND1 (Cyclin D1) by promoting its K63-linked polyubiquitination, thereby driving tumor cell proliferation and metastasis[2]. Germline mutations or loss of FBXO43 function can cause infertility due to disrupted meiosis. As of the latest information, no approved drugs directly target FBXO43, but it is considered a promising candidate for future molecular-targeted therapies in oncology and reproductive medicine[1][2][3].
