Overview

F-box only protein 47 (FBXO47) is a meiotic F-box protein that belongs to the SCF (Skp1–Cullin1–F-box) E3 ubiquitin ligase complex family, functioning in the regulation of meiotic chromosome dynamics. FBXO47 is highly expressed in the testis and plays a critical role in male meiosis, specifically in mediating the attachment of telomeres to the inner nuclear envelope, facilitating homologue pairing, and maintaining proper centromere pairing and synapsis during prophase I. FBXO47 interacts with key meiotic proteins such as SKP1 and members of the telomeric shelterin complex (TRF2, TRF1), stabilizing these proteins and preventing their degradation, which is essential for telomere function and proper chromosome segregation. Disruption of Fbxo47 in mice results in defective synapsis, impaired double-strand break repair, failure of meiotic recombination, and male infertility. There are currently no drugs that target FBXO47, and it is not considered a clinical therapeutic target.
