Overview

F-box only protein 50 (FBXO50, formerly known as NCCRP1) is a member of the F-box protein family involved in E3 ubiquitin ligase complexes, with a recognized F-box-associated (FBA) domain and evolutionary relationships to lectin-type F-box proteins[1][2][5]. FBXO50 is predominantly cytosolic and is implicated in protein-protein interactions that facilitate the ubiquitination and subsequent proteasomal degradation of specific substrates, particularly glycoproteins, although its lectin activity has yet to be experimentally confirmed[1][2][3]. In fish and mammals, the protein is expressed in nonspecific cytotoxic cells, where it is thought to contribute to their cytolytic activity[1][2]. Its expression is upregulated in specific pathological contexts, most notably in carbonic anhydrase IX-deficient mice, suggesting roles in cell proliferation and possibly tumorigenesis[1][3]. FBXO50 contains an FBA domain and shares homology with other FBXO family lectin-type proteins but possesses a structurally truncated F-box domain in mammals compared to canonical members[1][2][5]. No currently approved drugs or clinical biomarkers are reported that target FBXO50 directly.
