Overview

F-box protein 11 (FBXO11) is a member of the F-box protein family, characterized by an F-box domain which allows it to serve as a substrate recognition subunit of the SKP1–Cullin–F-box (SCF) E3 ubiquitin ligase complex. This protein promotes ubiquitination and proteasomal degradation of numerous regulatory proteins, including BCL6, SNAIL, CDT2, CIITA, and p53, thereby modulating processes such as cell cycle progression, immune regulation, epithelial–mesenchymal transition, tumor suppression, and hematopoietic cell fate. FBXO11 exhibits additional enzymatic activity as a protein arginine N-methyltransferase. Its dysregulation is implicated in various malignancies, neurodevelopmental disorders, immune diseases such as vitiligo, and chronic middle ear infections. FBXO11 expression can serve as a biomarker for prognosis and immune function due to its specific regulation of MHC class II expression in immune cells.
