Overview

F-box protein 15 (FBXO15) is a member of the F-box protein family, which are defined by a 40-amino acid F-box motif that mediates interaction with SKP1 as a part of SCF (Skp1-Cullin-F-box) E3 ubiquitin ligase complexes[1][3][4]. FBXO15 acts as a substrate-recognition component, enabling ubiquitination and subsequent proteasomal degradation of target proteins, including P-glycoprotein (ABCB1), thereby affecting protein levels that modulate multidrug resistance in cancer cells[1]. In stem cell biology, FBXO15 is transcriptionally activated by pluripotency factors such as Oct3/4 and Sox2, and is predominantly expressed in undifferentiated embryonic stem cells[1][3]. Genetic studies indicate FBXO15 is *not essential* for embryonic stem cell self-renewal or embryonic development, but its regulated expression highlights a functional role in pluripotency networks and cellular protein turnover. To date, FBXO15 is not considered a direct therapeutic target, but it participates in cellular processes relevant to cancer biology and stem cell maintenance[1][3][4][5].
