Overview

F-box protein 34 (FBXO34) is a member of the F-box protein family characterized by a conserved 40-amino acid F-box motif[1]. It serves as a substrate recognition component of the SCF (Skp1–Cullin–F-box) E3 ubiquitin ligase complexes, thereby regulating proteasomal degradation of proteins involved in key cellular processes, particularly during the cell cycle[1][3]. FBXO34 has been shown to play a pivotal role in oocyte meiotic maturation: loss of FBXO34 function leads to impaired G2/M transition due to insufficient maturation-promoting factor (MPF) activity, and overexpression disrupts anaphase entry due to continuous activation of the spindle assembly checkpoint (SAC), resulting in metaphase I arrest[1][3]. While F-box proteins as a family have documented links to cancer, diabetes, and germ cell development, specific links between FBXO34 and human disease remain limited in the literature, and no drugs or established biomarkers target this protein as of current knowledge[1][3][2].
