F-box protein 40 is a component of the SCF-type E3 ubiquitin ligase complex and is encoded by the FBXO40 gene in humans. It contains an approximately 40 amino acid F-box motif that mediates interaction with the SKP1 protein, enabling assembly of the SCF complex. FBXO40 serves as a substrate-recognition component, promoting the ubiquitination and subsequent degradation of specific protein targets, most notably insulin receptor substrate 1 (IRS1), which modulates the insulin-like growth factor 1 (IGF1) signaling pathway. Through this mechanism, FBXO40 can limit downstream IGF1 activity and may influence muscle and neuronal biology. Although the roles of F-box protein 40 in cancer and neurodegenerative disorders have been proposed based on broader family function, there is limited direct evidence linking FBXO40 to specific diseases, and it is not currently a validated therapeutic or diagnostic target.
