Overview

The F77 carbohydrate antigen is a tumor-associated glycan specifically overexpressed in prostate cancer cells, including both primary tumors and metastatic lesions (Zhang et al., 2010, PNAS). It is characterized as a branched I-type carbohydrate chain, specifically a poly-N-acetyllactosamine structure (Gao et al., 2014, J. Biol. Chem.). While its expression is high in malignant prostate tissue, it is significantly restricted in normal human tissues, making it a promising target for targeted therapies (Nonaka et al., 2013, Glycobiology). The monoclonal antibody F77 (mAb F77) was developed to recognize this antigen and has demonstrated the ability to induce cell death through antibody-dependent cellular cytotoxicity (ADCC) and complement-dependent cytotoxicity (CDC) in preclinical models (Zhang et al., 2010, PNAS). Research suggests that targeting the F77 antigen could provide a novel approach for treating androgen-independent and metastatic prostate cancer (Hatano et al., 2017, Biological and Pharmaceutical Bulletin).

Mechanism of action

The primary mechanism of action involves the use of monoclonal antibodies (e.g., mAb F77) to bind the antigen on the surface of prostate cancer cells, subsequently triggering antibody-dependent cellular cytotoxicity (ADCC) and complement-dependent cytotoxicity (CDC) (Zhang et al., 2010, PNAS).
