The Facilitates Chromatin Transcription (FACT) complex is a heterodimeric protein complex consisting of SUPT16H and SSRP1. It functions as a histone chaperone, playing a crucial role in chromatin remodeling during transcription, DNA replication, and DNA repair. FACT destabilizes nucleosomes to facilitate the passage of RNA polymerase II and other enzymes through chromatinized DNA templates. Overexpression of FACT is associated with aggressive cancers, while defects can lead to intellectual disability syndromes.
