Overview
Factor XII (FXII), also known as Hageman factor, is a plasma glycoprotein and a zymogen (inactive precursor) of the serine protease FXIIa. It plays a key role in the initiation of the intrinsic pathway of blood coagulation and is involved in several physiological and pathological processes, including thrombosis, inflammation, and bradykinin-mediated angioedema. FXII inhibitors are under investigation as novel anticoagulants because they may prevent pathologic clotting without increasing bleeding risk—a major limitation of current therapies targeting other factors like thrombin or factor Xa.
