FAM111A divergent transcript (FAM111A-DT) is a long noncoding RNA (lncRNA) transcribed from the genomic region near the FAM111A gene. Unlike the protein-coding FAM111A, FAM111A-DT does not encode a protein. Instead, it functions in the regulation of gene expression. Recent research has indicated that FAM111A-DT is upregulated in hepatocellular carcinoma (HCC), where it facilitates cancer cell proliferation, DNA replication, and tumor growth. These functions are mediated through N^6-methyladenosine (m^6A) modification, which enables FAM111A-DT to bind the m^6A reader YTHDC1 and recruit the histone demethylase KDM3B, thereby activating the FAM111A promoter and increasing FAM111A expression. FAM111A-DT has been suggested as a negative prognostic marker in HCC and is a candidate for future therapeutic targeting, although it currently has no approved drugs targeting it directly.
