Overview

FAM151B-DT is a cytoplasmic long non-coding RNA located on chromosome 5q12.1, transcribed from the negative strand in divergent orientation. It is widely expressed in the central nervous system, especially in neurons. This lncRNA modulates autophagic flux by acting as a scaffold or mediator for chaperone proteins like HSC70, facilitating the lysosomal degradation of proteins prone to aggregation, specifically tau and α-synuclein. Silencing FAM151B-DT blocks autophagy, leading to pathologic protein accumulation, while increasing its expression promotes autophagic clearance and reduces neurotoxic protein aggregates. FAM151B-DT is altered in stem cell models of neurodegeneration and in human brains afflicted with Alzheimer’s disease, Parkinson’s disease, FTLD-tau, and PSP. This molecular profile and functional data designate FAM151B-DT as a novel and promising therapeutic target for neurodegenerative diseases.
