Overview

FAM169A antisense RNA 1 (FAM169A-AS1) is a long non-coding RNA transcribed in the antisense orientation to the FAM169A gene. It is classified with the lncRNAs and is not a conventional pharmaceutical target (such as a protein receptor or enzyme). However, its circular RNA isoform, circ-FAM169A, has been shown to modulate extracellular matrix homeostasis in degenerative disc cells by acting as a microRNA sponge. Increased expression of circ-FAM169A in degenerative disc disease promotes matrix degradation via the NF-κB pathway. FAM169A-AS1 also appears as a candidate genomic locus associated with protein metabolism in livestock, reflecting its broader regulatory potential in gene expression[1][2][4]. There are currently no drugs that target FAM169A-AS1, and its clinical application is investigational. Its biological activities suggest it operates by regulating RNA-mediated pathways rather than acting as a classical therapeutic target.

Mechanism of action

Not a therapeutic target itself, but *its modulation* (particularly via antisense oligonucleotides or siRNAs) could theoretically alter disease pathways by affecting the expression of its target RNAs or interacting microRNAs. Its circular RNA isoform (*circ-FAM169A*) acts as a molecular sponge to sequestrate miR-583, indirectly relieving repression of targets such as **BTRC** and **Sox9**, thereby affecting NF-κB signaling and cell survival pathways.
