FAM83A antisense RNA 1 (FAM83A-AS1) is a long non-coding RNA (lncRNA) transcribed antisense to the FAM83A oncogene, located on chromosome 8q24.13[2][3]. FAM83A-AS1 is upregulated in several cancers, especially non-small cell lung cancer (NSCLC) and lung adenocarcinoma, where its high expression is associated with poorer patient survival[1][2]. Functional studies demonstrate that silencing FAM83A-AS1 suppresses cancer cell proliferation, migration, invasion, and colony formation, while inducing cell cycle arrest and autophagy[1]. Mechanistically, FAM83A-AS1 increases the stability of FAM83A pre-mRNA (by forming an RNA duplex and recruiting RNA-binding proteins), thereby upregulating FAM83A expression and promoting oncogenic pathways such as MET/EGFR and ERK signaling[1][2]. FAM83A-AS1 knockdown downregulates multiple cancer- and signaling-related genes, and has emerged as a putative therapeutic target and biomarker for cancer diagnosis, prognosis, and potentially therapy response[1][2]. No approved drugs currently target FAM83A-AS1 directly, but it is being explored as a target for oligonucleotide-based therapeutics.
