Family with sequence similarity 110 member A (FAM110A) is a protein localized to centrosomes and spindle poles with cell cycle–regulated expression. FAM110A is critical in mitotic progression, promoting organization of the mitotic spindle by binding and crosslinking actin (N-terminal domain) and tubulin (C-terminal domain), facilitating proper chromosome alignment, spindle assembly, and metaphase-to-anaphase transition. Its phosphorylation by Casein Kinase I is crucial for its function in mitosis[1][5][7]. It accumulates at the microtubule-organizing center during interphase and spindle poles during mitosis[1][3][7]. FAM110A is not a traditional therapeutic target (such as a receptor, enzyme, or transporter) but has emerged as a potential biomarker in cancer prognosis, particularly in breast cancer and several other malignancies, as well as being implicated in neuropsychiatric and cardiovascular conditions[1][6]. Direct drug interactions or therapeutic strategies targeting FAM110A have not been reported in the literature to date.
