Overview

Family with sequence similarity 110 member B (FAM110B) encodes a centrosome- and spindle pole–associated protein with critical roles in regulating cell cycle progression, particularly at the G1 phase. In non-cancer contexts, proper FAM110B expression supports centrosome function, while aberrant or ectopic expression can impair cell cycle progression. In cancer, FAM110B displays both oncogenic and tumor-suppressive functions depending on cellular context: it promotes tumor progression in castration-resistant prostate cancer (via androgen receptor signaling) but restricts proliferation and invasion in non–small cell lung cancer (by inhibiting Wnt/β-catenin signaling). FAM110B also modulates tumor microenvironment characteristics by regulating immune cell infiltration and inflammation, and it is under investigation as a prognostic biomarker and a potential therapeutic target for immunotherapy in diverse cancers. Its biological function extends further to processes such as DNA repair, apoptosis, angiogenesis, and stemness, as well as transcriptomic effects mediated through epigenetic modifications.
