Family with sequence similarity 120 member B (FAM120B) is a protein-coding gene that encodes a transcriptional coactivator involved in the regulation of transcriptional programs mediated by peroxisome proliferator-activated receptor gamma (PPARG) and estrogen receptor 1 (ESR1)[2][3][6]. FAM120B acts as a constitutive coactivator for PPARG, promoting adipogenesis (the formation of fat cells) through the activation of PPARG pathways, and is predicted to play a role in fat cell differentiation and metabolism[1][2][3][6]. The protein is mainly localized to the nucleus. Functional studies also link FAM120B to immune regulation and reproductive biology, such as germ cell development, and it may modulate disease susceptibility, including in metabolic and infectious diseases[1]. No drugs directly targeting FAM120B are currently known, nor are FAM120B-specific biomarkers or safety concerns described. However, its role as a coactivator of PPARG situates it as a participant in pathways known to be targeted by anti-diabetic and metabolic drugs, although FAM120B itself is not a direct therapeutic target[3][4][5].
