Family with sequence similarity 131 member A (FAM131A) is a protein encoded by the FAM131A gene in humans, located on chromosome 3q27.1[1]. The gene produces a nuclear protein, mainly localized to the nucleoli rim, with two known isoforms and predicted post-translational modifications such as sumoylation and lysine acetylation[1]. FAM131A is highly expressed in the brain, but shows overall low tissue specificity[1]. Functional characterization remains limited, but overexpression is associated with adverse prognosis in ovarian and endometrial cancers[1]. Additionally, FAM131A is implicated in a FAM131A–BRAF gene fusion that may contribute to oncogenic signaling (MAPK pathway activation) in rare aggressive prostate cancer variants, suggesting its potential as a cancer biomarker[3]. No drugs are currently known to directly target FAM131A, and its physiological role is not fully defined.
