Family with sequence similarity 135 member B (FAM135B) encodes a large protein of unknown definitive function, with domains DUF676 (putative serine esterase) and DUF3657, expressed in various healthy tissues including brain and pancreas, and more strongly in certain tumors. Its high expression in ESCC tumors correlates with advanced stage, lymph node metastasis, and radioresistance, acting as a context-dependent promoter of cell proliferation and survival via the PI3K/Akt/mTOR pathway. In contrast, reduced FAM135B is implicated in neurodegenerative phenotypes. Genetic variation in FAM135B may also affect immune response and susceptibility to infectious disease, such as tuberculosis. Although FAM135B has no direct interacting drugs, its expression levels serve as a potential biomarker for cancer progression and treatment response
