Family with sequence similarity 149 member A (FAM149A) is a conserved human protein comprised of 482 amino acids, encoded by the FAM149A gene located on chromosome 4q35.1. It contains a single characterized domain of unknown function (DUF3719) and has no confirmed enzymatic or receptor activity, nor defined roles in cell signaling, membrane transport, or transcription. FAM149A exhibits high sequence conservation across mammals and other vertebrates, and is expressed in a variety of human tissues, with prominent levels in the trigeminal ganglion, kidney, pancreas, adrenal gland, and moderate levels in the heart and throughout most of the body. Its function is not fully understood, but limited evidence suggests potential involvement in cellular responses to oxidative stress through pathways such as Nrf2-antioxidant signaling. FAM149A may play a role as a candidate gene in the development of certain cancers and dysplastic lesions, with decreased expression reported in oral cancer. No established small-molecule drugs, therapeutic antibodies, or clinical biomarkers specifically target or utilize FAM149A at present, and its relevance as a therapeutic target remains unproven. There are also no reported safety concerns or adverse phenotypes associated with its modulation[1][3][4].
