Family with sequence similarity 151 member A (FAM151A)

Target
FAM151A
Molecular classification
Transmembrane protein, Member of PLC-like phosphodiesterase superfamily, Contains DUF2181 domains (Domain of Unknown Function 2181), which belongs to the GDPD/PLCD superfamily
01

Overview

FAM151A is a protein-coding gene encoding a transmembrane protein mainly expressed in kidney tubules in humans. It shares homology with menorin, a nematode protein essential for dendrite branching and neuronal development. The mammalian ortholog likely contributes to neuronal connectivity, although knockout models in mice show no overt phenotype for FAM151A itself (distinct from its paralog FAM151B, which is essential for retinal function in mice). The protein contains DUF2181 domains, with partial predicted phosphodiesterase activity, but its substrates and precise function remain undefined. Variation in FAM151A (notably SNP rs11206394) has been linked to reduced risk of colorectal cancer, suggesting a role as a genetic biomarker rather than a therapeutic target. FAM151A is found in extracellular exosomes and is associated with several molecular functions through genetic, transcript, and proteomic analyses, but its direct functional roles and druggability remain uncharacterized.

Mechanism of action

Not applicable. No drugs are known to act on FAM151A.

03

Biological functions

Putative involvement in sensory dendrite morphogenesis (inferred from homology to C. elegans menorin, which mediates neuronal connectivity)Presumed phosphodiesterase-like function (by family/domain annotation), though specific biochemical activity and substrates remain undefined
04

Disease associations

Colorectal adenocarcinoma: FAM151A SNP rs11206394 is a significant predictor, with certain alleles linked to reduced riskAutism spectrum disorder: gene association reported, but mechanism and relevance remain unclear
05

Safety considerations

None documented. The gene does not present notable therapeutic challenges as a drug target; implications in disease relate to genetic variation rather than therapeutic intervention
06

Biomarkers

SNP rs11206394: genetic biomarker predicting colorectal cancer riskExpression in extracellular exosomes: occasionally reported in diagnostic context
