Overview

Family with sequence similarity 155 member B (FAM155B, formerly NALCN channel auxiliary factor 2 or NALF2) is a transmembrane protein that serves as an auxiliary subunit in the NALCN sodium leak channel complex. It is essential for proper membrane localization and assembly of the NALCN complex and, along with other subunits (UNC79, UNC80), regulates neuronal excitability by maintaining resting membrane potential[2][4]. FAM155B is a cysteine-rich domain protein that participates in stabilizing and trafficking the pore-forming NALCN channel subunit to the plasma membrane. Genetic disruptions in NALCN or its auxiliary components, including FAM155 proteins, are implicated in neurodevelopmental disorders due to impaired channel function[1][4]. There are no established direct pharmacological modulators or biomarker roles for FAM155B at this time.
