Family with sequence similarity 163 member A (FAM163A) is a secretory protein originally identified in neuroblastoma cell lines and predominantly expressed in the brain. It has roles in promoting cell proliferation—particularly in cancer—through direct interaction with ERK and 14-3-3β, activating the ERK signaling pathway. FAM163A is also involved in the regulation of mitochondrial dynamics and biogenesis in neurons, affecting hypothalamic neuron activity tied to metabolic control. Overexpression of FAM163A is associated with poor prognosis in some cancers, such as lung squamous cell carcinoma. Its biological functions remain incompletely characterized, but current research suggests relevance in cancer biology, neuronal metabolism, and mitochondrial regulation[1][2].
Drugs such as MEK inhibitors (U0126) act by blocking the phosphorylation of ERK and downstream signaling potentiated by FAM163A[2].
