Family with sequence similarity 168 member A (FAM168A, also known as TCRP1 and KIAA0280) is a protein-coding gene implicated in oncogenesis and cancer therapy resistance. It functions primarily as a regulator of base-excision DNA repair and protects cells from DNA damage-induced apoptosis. In cancer cells, FAM168A acts as a linker protein by directly interacting with BCR-ABL1 and AKT1, promoting AKT1 phosphorylation and activating the NFκB pathway, which enhances cell survival and proliferation. FAM168A also prevents degradation of POLB, stabilizing its protein levels. Overexpression of FAM168A is linked to chemotherapy resistance, particularly in tongue cancer and oral squamous cell carcinoma, and has been identified in fusion genes in several cancer types.
Modulation of chemotherapy resistance via regulation of DNA repair and inhibition of apoptosis pathways (PI3K/AKT/NFKB). By acting as a linker protein, FAM168A enhances AKT1 phosphorylation, modulates NFκB signaling, and influences cell survival and proliferation.
