FAM170B is a testis-expressed structural protein, localized mainly to the acrosomal vesicle and perinuclear region of spermatids and spermatozoa[2]. It is involved in the acrosome reaction, a process critical for fertilization in mammals, likely through interactions with GOPC (Golgi-associated PDZ and coiled-coil motif-containing protein) during acrosome biogenesis[2][6][12]. While experimental inhibition of FAM170B impairs fertilization in vitro in mice, current evidence indicates it acts as an acrosomal participant rather than a classical therapeutic target, receptor, or enzyme[2][7][9]. Human gene databases annotate its activity as “positive regulation of acrosome reaction” and “regulation of fertilization,” with primary localization to sperm acrosomes[1][8][12]. No clinically relevant drug interactions, biomarker status, or disease links have been described; FAM170B thus remains a structurally and biologically defined fertility-related protein, not a recognized drug target at present.
