Its expression is cyclic in the presomitic mesoderm of murine embryos in phase with Notch-regulated targets, suggesting a role in regulating somitogenesis. FAM181B directly interacts with TEAD transcription factors via an Ω-loop and a conserved LxxLF motif, indicating a regulatory function within the Hippo pathway that may influence transcriptional programs required for tissue development, particularly in the nervous system. While it has emerged in some association studies (e.g., with growth traits in chickens), functional disruptions do not produce obvious phenotypes in mice, potentially due to redundancy with the related protein FAM181A.
