Family with sequence similarity 220 member A (FAM220A) encodes a protein that interacts with STAT3 and promotes its dephosphorylation, thereby negatively regulating STAT3 transcriptional activity. FAM220A is expressed intracellularly, predominantly within the nucleus and acrosomal vesicle. Despite its widely conserved sequence and multiple aliases, its precise physiological functions and disease associations are still largely uncharacterized; current evidence points mainly to its role as a modulator of STAT signaling through protein-protein interaction and regulation of phosphatase activity. There is no established therapeutic targeting, associated drugs, or biomarker utility for FAM220A at present.
