Family with sequence similarity 221 member A (FAM221A) is a protein-coding gene located on chromosome 7p15.3 in humans, encoding a 298 amino acid protein with a molecular weight of approximately 33.1 kDa[2][12]. FAM221A is expressed in several tissues, with higher expression in the spinal cord, pancreas, retina, liver, brain, fetal brain, thyroid, and colon[2][8]. There is emerging evidence linking FAM221A to diseases such as Parkinson’s disease, Alzheimer’s disease, prostate cancer, and colorectal tumors, though its precise molecular function remains uncharacterized[1][2]. It has one paralog, FAM221B, and interacts with proteins involved in intracellular trafficking, notably SNX2, SNX5, and SNX6[2]. No drugs are currently known to target FAM221A, and it is not established as a therapeutic target[1][2][12].
