The protein is highly expressed in the testis, kidney, adrenal, and thyroid, with lower expression in many other tissues. It contains domains of unknown function and is predicted to be localized to the nucleus, possessing multiple nuclear localization signals and a vacuolar targeting motif. While its precise biological role is not yet characterized, evidence links FAM227B to differential methylation in fertility, cancer risk loci for oral, breast, and lung cancers, and differential expression in cardiovascular disease. The gene participates in fusion events (e.g., SUGP2-FAM227B) associated with pan-cancer fusion gene contexts. There are currently no drugs or direct mechanisms modulating FAM227B, making it primarily an investigational protein with clinical correlations but not a recognized therapeutic target.
