Overview

Family with sequence similarity 237 member A (FAM237A) is a small secreted peptide and protein, primarily expressed in the human brain, encoded by the FAM237A gene[2][5]. The canonical active form is a C-terminally amidated peptide containing 80 amino acids and a disulfide bond, required for its biological activity[1][4][6]. FAM237A functions as the endogenous agonist of the GPR83 receptor, binding with high affinity and specificity and activating signaling via the GNAQ pathway[3][4][6]. Multiple isoforms are predicted, but the physiologically relevant species is the amidated, processed form starting from residue 34, forming a ~9 kDa peptide[6]. FAM237A may play a role in neuropeptide signaling and central nervous system processes through GPR83 activation; however, its involvement in specific disease states or as a biomarker has not been demonstrated[3][6]. There are currently no reported drugs interacting with FAM237A, nor direct safety or therapeutic challenges described in the literature[5][6].
