Overview

Family with sequence similarity 3, member D antisense RNA 1 (FAM3D-AS1) is a conserved long non-coding RNA (lncRNA), predominantly localized in the cytoplasm, which does not encode a protein. It plays a regulatory role in cellular processes such as inhibition of cell proliferation, invasion, and epithelial-mesenchymal transition (EMT), particularly in colorectal cancer. Overexpression of FAM3D-AS1 inhibits tumor-associated behaviors and may reverse EMT via modulation of the NF-κB signaling pathway. There is emerging evidence for its function as a tumor suppressor lncRNA, with decreased expression in colorectal cancer tissues compared to adjacent normal tissues. Its potential relevance in other diseases, such as endometrial cancer, is under study, though there is currently no strong evidence linking it as a risk factor in that context. FAM3D-AS1 is not currently classified as a druggable therapeutic target (like a receptor, enzyme, or transporter), nor are there clinically approved drugs targeting it directly, but it is being explored as a candidate biomarker and novel regulator in cancer biology[1][2][4][5][6].
