Overview

Family with sequence similarity 30 member A (FAM30A) is a long non-coding RNA (lncRNA), over 200 nucleotides in length, and does not encode a protein[3][4][5]. It is primarily expressed in the spleen, KG-1 cell line, and peripheral blood[6]. FAM30A acts as a tumor suppressor in multiple cancers, with well-characterized roles in colorectal carcinoma where its low expression is associated with advanced disease, poor differentiation, and poor prognosis[1][2][4]. Mechanistically, FAM30A inhibits cell proliferation, migration, and metastasis, and promotes apoptosis in colorectal cancer cells, at least partly by negatively regulating the JAK–STAT signaling pathway[1]. Additional evidence links FAM30A to the regulation of immune cell infiltration, with correlations to B cells, CD4+ T cells, and dendritic cells, and involvement in B-cell and T-cell receptor pathways[1]. It also acts as a molecular sponge for specific microRNAs such as miR-21-3p, further impacting tumor progression[2][4]. FAM30A is underinvestigated as a direct therapeutic target—no drugs are known to target FAM30A directly—and its primary utility at present is as a novel prognostic biomarker for colorectal carcinoma and potentially other malignancies[1][2][4].
