Family with sequence similarity 53 member A (*FAM53A*) encodes a protein initially linked to neural development, particularly the specification of dorsal neural tube cell fate[1][2][3]. Recent research has revealed its involvement in cancer cell proliferation, migration, and invasion, with effects highly dependent on p53 tumor suppressor status: in p53 wild-type cells, FAM53A acts as a tumor suppressor by inhibiting MEK/ERK signaling and related cell growth and migration, whereas in p53-deficient cells, it can promote tumor progression by increasing MEK/ERK activity[2]. Genetics studies also associate FAM53A with Wolf-Hirschhorn syndrome and Blepharochalasis[1]. While FAM53A is not a classical drug target such as a receptor or kinase, its modulation of critical signaling pathways places it among emerging candidate targets for cancer therapy, biomarker development, and disease prognostication[2][10].
