Overview

Family with sequence similarity 53 member B (FAM53B) is a protein-coding gene and member of the FAM53 family involved in the regulation of the Wnt signaling pathway, primarily by modulating the nuclear localization of beta-catenin (CTNNB1)[1][3][6][7]. FAM53B binds to 14-3-3 chaperones and Ski-interacting proteins (SKIIP), the latter being spliceosome-associated proteins that participate in the regulation of transcription[2]. Expression of FAM53B is detected in several human tissues, including skeletal muscle, kidney, spleen, thyroid, testis, ovary, small intestine, colon, peripheral blood, and various regions of the brain[1][11]. Functionally, FAM53B has been shown to play a regulatory role in cell proliferation, neural tube development, and, in cancer models, the promotion of tumor metastasis by enhancing macrophage M2 polarization, especially in pancreatic ductal adenocarcinoma (PDAC)[4][10]. Chromosomal translocations leading to FAM53B truncation have been detected in acute lymphoblastic leukemia, with evidence suggesting a potential tumor suppressor role via the Wnt/β-catenin pathway[2]. As of now, there are no known drugs targeting FAM53B, and it is not established as a druggable or pharmacologically actionable therapeutic target.
